Poorly water-soluble active pharmaceutical ingredients frequently demand more than a conventional solubility-enhancement approach. An amorphous solid dispersion can increase apparent solubility and accelerate dissolution, but its performance depends on a tightly connected set of decisions: drug–polymer compatibility, polymer concentration, manufacturing route, moisture exposure, and what happens when the formulation meets gastrointestinal fluids.

For R&D teams, the difficult questions begin after amorphization. A dispersion may show no detectable crystalline peaks by X-ray powder diffraction yet produce inadequate supersaturation, phase separation or inconsistent drug release. Increasing drug loading may improve dosage-form feasibility but reduce stability or precipitation resistance.

Similarly, spray drying and hot-melt extrusion can create different particle attributes, thermal histories and downstream processing demands, even with the same drug and polymer.

This training examines how to evaluate these using formulation science, process understanding and meaningful performance data. Participants will compare selection criteria for HPMCAS, copovidone, HPMC and other functional carriers; assess spray-drying and extrusion variables; interpret solid-state and dissolution results; and identify risks during storage, milling and tableting. Technical examples will connect analytical findings to formulation decisions, helping participants select, troubleshoot and qualify amorphous solid dispersions with greater confidence.

The emphasis is on technical decisions that can be defended with evidence, not simply producing an amorphous powder.

What Problem This Training Solves

  • Uncertain polymer selection: Choosing a carrier that maintains drug–polymer compatibility while supporting dissolution and physical stability.

  • High drug-loading limitations: Increasing API content without sacrificing supersaturation, release performance or manufacturability.

  • Unclear process selection: Deciding between spray drying and hot-melt extrusion using material properties and manufacturing constraints.

  • Misleading analytical results: Understanding why apparently amorphous samples may exhibit poor dissolution or hidden phase separation.

  • Unpredictable stability: Evaluating crystallization, moisture sensitivity and performance changes during processing and storage.

  • Downstream formulation difficulties: Maintaining ASD performance through milling, granulation, compression and final dosage-form development.

Why Attend?

Participants will gain practical capabilities to:

  • Compare polymer carriers using drug-specific physicochemical properties and performance requirements.

  • Make better-informed spray-drying and extrusion decisions using meaningful processing variables.

  • Interpret characterization and dissolution results together rather than relying on individual analytical tests.

  • Evaluate high-drug-loading formulations against dissolution, stability and dosage-form constraints.

  • Apply practical troubleshooting methods to realistic ASD development scenarios.

  • Prepare more defensible experimental plans and technical development decisions.
